brscFSH (Temporarily Out of Stock)

$140.00 USD

Regulatory Status: RUO

Other Names: Follicle stimulating hormone (or FSH), gonadotrophic hormone.

Description: FSH is a glycoprotein polypeptide hormone with alpha and beta subunits that plays a crucial role in the reproductive system of both males and females. It is synthesized and secreted by the gonadotropic cells of the anterior pituitary gland in response to gonadotropin-releasing hormone (GnRH) from the hypothalamus.

The bscFSH hormone is a single-chain variant of bovine follicle-stimulating hormone (FSH), designed by fusing the beta and alpha subunits through a 33-amino acid glycine- and serine-rich spacer peptide derived from human keratin 10. This structure includes two additional N-glycosylation sites in the internal portion of the spacer to improve its stability and functionality. Additionally, a six-residue histidine tag has been incorporated at the C-terminal end to facilitate detection and purification.

A detailed description of the sequence of this molecule can be found in the publication Development and characterization of a novel variant of long-acting bovine follicle-stimulating hormone (bscFSH) (Theriogenology, 2024, doi: 10.1016/j.theriogenology.2024.05.038).

Molecular Mass: The mature molecule consists of a total of 247 amino acids with a predicted molecular weight of 26.4 kDa. However, it also contains six potential N-glycosylation sites occupied by bi- and tri-antennary chains, adding an additional weight of 20 to 26 kDa. As a result, in a 12% SDS-PAGE under reducing conditions, the molecule migrates as a single diffuse band with an average molecular weight of approximately 48 kDa.

Purity: > 97% by SDS-PAGE gel as determined by Coomassie stained SDS-PAGE.

Formulation: The product is supplied as a lyophilized powder, with each vial containing 300 µg of FSH. The protein was lyophilized from 1 mL of a 0.22 µm filtered solution, formulated with sucrose (49.37 g/L), sodium citrate (14.3 g/L), methionine (0.7 g/L), and Tween 20 (0.02%), at pH 7.0.

Concentration: Each vial contains 300 µg of lyophilized FSH, which must be reconstituted in the 20 mL vial of reconstitution solution provided. This results in a final solution with a concentration of 15 µg/mL of FSH. A dose and administration schedule recommendation table is included in the package insert to guide appropriate usage.

Storage & Handling: The hormone is distributed at ambient temperature (25°C). Upon receipt, it should be stored at 2°C to 8°C for up to two years. Once reconstituted, the product has been shown to retain its activity for up to two months if kept at 4°C.

Application: Bioassay
